Background Info: Lydia Elsher is the young sister of Kaitlynn Elsher and the youngest daughter of Keith and Betty Elsher.

Lydia and her older sister, Kaitlynn join their family friend, April O'Neil and her turtle friends, Leo, Raph, Donnie, and Mikey to stop an army of mutants that have been unleashed. 

In the present after 15 years later of a lab destruction, 23 year old college student Lydia Elsher is raised by her parents, Keith and Betty Elsher with the help of her 33 year older sister, Kaithlyn Elsher to defend herself and get home on time while four turtle brothers, Mikey, Leo, Raph, and Donnie were raised, taught the arts of ninjitsu, and were instructed to only leave for the surface world to get supplies for their home in the sewer as told by their adoptive father, Splinter.

After her secret to stay out past her curfew, Keith tells Lydia a backstory about how he got the scratch scars and when Lydia was just a Middle Schooler, she and Keith were out for a father-daughter bonding time when a group of mutants jumped them both and scratched up Keith, who uses himself as a human shield in front of Lydia, resulting in Keith getting Lydia into martial arts, karate, and ninjitsu to defend herself against anyone who would hurt her.

Sometime later on during one of her nights off from the pizza place, Lydia and the Elsher's family friend, April O'Neil get their attention the turtles as they were yelling at them while they were not seeing them in the shadows, April gets her moped stolen and Lydia gets her stone necklace stolen as well.

The Turtles are able to get those two items back after defeating the group that stole them and revealed themselves to them, resulting Lydia to call up Kaitlyn excitedly about the Turtles.

Lydia, Kaitlyn, and April show the turtles the high school where the group discusses their goals, the turtles want to be embraced by the surface world while Lydia dreams of graduating college to become a Zoologist while finding someone who she didn't care how they looked and April is an aspiring journalist, who wants to move past an embarrassing viral incident where she vomited from anxiety during her school announcements.

The six young adults decide to help each other out; Lydia and April have been investigating a series of violent robberies on TCRI technology by a criminal named Superfly, which have led parents to call for a city-wide curfew that would cancel the school's Prom Lydia and Kaitlyn are help chaperoning on.

If April records and writes an article on who Superfly is and how the Turtles and Lydia would stop him, April, Lydia, and the Turtles would be regarded as heroes and accepted.

Using leads from April and information from Lydia, the Turtles along with Lydia's help, find out that Superfly is collecting a piece of technology that night in a deal under the Brooklyn Bridge.

The four turtle brothers and Lydia are able to find the part and pose as buyers and discover that not only is Superfly a fly mutant, but also has a gang of mutants with him like Leatherhead, Mondo Gecko, Genghis Frog, Wingnut, Scumbug, Ray Fillet, and Bebop and Rocksteady.

Soon, a secret romance between Lydia and Rocksteady starts to grow upon meeting each other during the meeting.

Ecstatic that there are other mutants, Superfly invites the Turtles and Lydia to go on a night in the town with him and his gang, where they bond.

Superfly explains his story and plan: like the Turtles, the mutagen evolved him and the rest of his mutant gang and he explains that Baxter Stockman took on the role of their father figure. Years later after the lab explosion and Baxter Stockman's possible death, when Superfly brought the Mutants to the surface, they too were nearly killed by never accept them, he would create a weapon that would turn the ooze into an airborne virus, which would mutate all non-human wildlife on the planet and make them the dominant species while enslaving humanity in the process.

The Turtles and Lydia try to stop them by trying to escape with the last part, but Superfly and his gang escape with it and Lydia as well and the Turtles are left to be captures by TCRI, who had a tracker on the last part.

Having followed the Turtles and Lydia from a distance, Kaitlyn and April leaves to get Splinter to rescue them.

Lydia is gets beaten up by Superfly at his lair and was going to bleed out from the beating while at TCRI, the Turtles are slowly "milked" of their mutagen before they are rescued by Splinter, Kaitlyn, and April.

Betrayed by his sons' lies and despite stating that Lydia, Kaitlyn, and April are the only good humans, Splinter decides that they will stop Superfly before they stay in the sewers permanently.

When they reach the island, a slowly healing Lydia, Leo, and Splinter plead to the conscious of Superfly's gang, stating that if they enslave humanity then they are no better than the human themselves.

The gang agrees and everyone fights Superfly, destroying the machine in the process.

Unfortunately, the ooze enlarges and combines Superfly with other local wildlife into a bipedal whale-like creature who dubs himself "SuperDuperFly", deciding to attack New York City.

The Turtles, Splinter, the Mutants, Lydia, Kaitlyn, and April head to mainland New York City to stop him once and for all.

While the Turtles, Splinter, the Mutants, Lydia, and Kaitlyn fight, April is able to overcome her anxiety and explain on live air to the public what's going on, leading the citizens of New York City to help stop SuperDuperFly.

Donnie discovers that the blowhole on the back of his neck carries a massive amount of mutagen and the Turtles, with Lydia's help, are able to drop a canister of anti-ooze into SuperDuperFly's blowhole, returning him to being a normal fly by using Leo's voice as a leader, utilizing Mikey's gift for improvisation, Donnie's intelligence, Raph's rage, and Lydia's animal knowledge and singing.

The Turtles, Splinter, Kaitlyn, April, Lydia, and the Mutants are recognized as heroes by the city and the boys and Lydia reconcile with their father and her parents, who apologizes for their xenophobic behaviors.

After reconciling with Keith and Betty, Lydia announces to her new friends that she has a secret crush on Rocksteady, who confesses his feelings to her as they both go into a relationship and they both kiss as their friends cheer for them and Keith goes into protective dad mode.

Soon after, the Mutants move into the sewers with the Turtles and Splinter with Lydia, Kaitlyn, Keith, Betty, and April visiting periodically and the Turtles enroll at April's high school with Lydia's help, where they are all embraced as heroes and with that, Splinter and Scumbug fall in love with each other. 

In a mid-credit scene, Lydia is shown now a graduate of college and the Turtles are shown enjoying high school life: Lydia is a fully licensed Zoologist to study different animals while still in a relationship with Rocksteady, Mikey is participating in the school's comedy improve team, Raph has joined the wrestling club, Donnie has found the computer lab, and Leo (who, throughout the film, has developed a crush on April) has decided to help her with her journalistic interests, now focused on TCRI who have mysteriously disappeared since the events of the film.

While the Turtles, Rocksteady, April, Lydia, and Kaitlyn are later enjoying themselves at the school prom, Cynthia, who is spying on them and with the now unmutated Superfly captive by her side, who plans to capture Lydia and recapturing the Turtles by enlisting the aid of the mysterious Shredder.
